ABSTRACT:
Modern pharmaceutical research increasingly depends on analytical technologies capable of generating orthogonal information on identity, purity, molecular structure, solid-state behavior, biological disposition and process performance. This review examines the role of modern analytical instrumentation across the drug-discovery and drug-development continuum, from hit identification and structural elucidation to preformulation, bioanalysis, stability assessment, manufacturing and quality control. Major platforms discussed include UV-visible, infrared, Raman and nuclear magnetic resonance spectroscopy; HPLC, UHPLC, GC and HPTLC; mass spectrometry and high-resolution MS; hyphenated systems such as LC-MS/MS and LC-HRMS; thermal analysis; X-ray diffraction; electron and atomic-force microscopy; particle-size and zeta-potential analysis; capillary electrophoresis; and ICP-based elemental analysis. Particular attention is given to the complementary nature of these techniques rather than treating instruments as isolated methods. Applications in natural-product drug discovery, metabolomics, impurity profiling, pharmacokinetics, nanopharmaceutical characterization and process analytical technology are highlighted. The review further considers analytical quality by design, chemometrics, artificial intelligence, automation, miniaturization, real-time release testing and green analytical chemistry. Current evidence indicates a shift from end-product testing toward integrated, high-information and increasingly predictive analytical workflows. The future analytical laboratory is likely to combine orthogonal instrumentation with automated sample handling, high-resolution data acquisition, multivariate modelling and machine learning while maintaining traceability, validation and regulatory control. Such integration can accelerate pharmaceutical decision-making, reduce analytical uncertainty and support the development and manufacture of safer, more consistent and clinically effective medicines.
